Hii Everyone, we are six people get off at Ajmer station by 3 am in morning and have to catch train by 08:30 PM from Ajmer itself on same day. We are planning for Srinathdwara temple darshan and pushkar darshan on same day. Is it possible? If yes then please suggest where to go first, and by which mode i.e taxi or bus. If it is taxi then what would be approximate cost??

Since you are getting off the train at Ajmer, it is obviously good to invest the day visiting Pushkar and Ajmer both. It will consume the entire day. Sri Nathdwara is close to Udaipur and in case you intend visit there you should go straight to Udaipur and not Ajmer. You simply cannot cover these places in just one day.
